A genetic change that becomes more common in men with age could help scientists identify some cancers at an earlier stage, according to new research. The study found that loss of the Y chromosome in cells may occur not only inside tumours but also in normal-looking tissue surrounding a tumour.
The findings published in the journal ‘JCI Insight’ suggest that loss of the Y chromosome could potentially act as an early warning sign of cancer development in men. However, more research is needed before it can be used as a routine cancer screening tool.
What Is Loss Of The Y Chromosome?
Men usually have one X and one Y chromosome in each cell. The Y chromosome carries genes involved in male biological development and other cellular functions.
As men get older, some cells can lose their Y chromosome. This is known as mosaic loss of the Y chromosome, or mLOY. It is a relatively common genetic change associated with ageing.
Previous research has linked loss of the Y chromosome in blood cells with an increased risk of certain diseases and poorer outcomes in some cancers. The new study looked more closely at whether this chromosome loss could also be detected in tissues before or around the time cancer develops.
Researchers Examined More Than 1,000 Tissue Samples
Scientists analysed 1,000 tissue samples from 405 men, covering 11 different organs. Using a technique that can identify X and Y chromosomes in individual cells, they examined more than 4.3 million cell nuclei.

The researchers compared normal tissue, tissue close to tumours, and cancerous tissue. They found that the amount of Y chromosome loss increased progressively across these stages.
The pattern was particularly noticeable in several cancers, including cancers of the colon, rectum, oesophagus, pancreas and lung. The highest degree of Y chromosome loss was seen in colorectal cancer.
Y Chromosome Loss: A Possible Early Sign Of Cancer
One of the most important findings was that Y chromosome loss was found in tissue that looked normal under a microscope but was located next to a tumour.
Researchers described this as a possible field effect. This means that genetic changes linked to cancer may extend beyond the tumour itself, creating an area of tissue that is more vulnerable to cancer development.
The researchers also created detailed maps of bladder tissue. These showed a gradual increase in Y chromosome loss from normal tissue to early abnormal changes and then to invasive cancer.
This gradual pattern could potentially help doctors identify early cancer-related changes in tissue samples, including situations where a biopsy does not capture a small tumour.

What does this mean for men?
The findings do not mean that losing the Y chromosome automatically causes cancer or that every man with Y chromosome loss will develop cancer.
Instead, the study suggests that Y chromosome loss may be part of the biological changes that occur as normal tissue moves towards cancer. Understanding this process could eventually help researchers develop new biomarkers for detecting cancer at an earlier stage.
The study also adds to previous research suggesting that loss of the Y chromosome may affect how cancer cells interact with the immune system.
